Hōrai Arc[]
After performing experimentations on Aza Chōbei, because of his unique Tao, Rien managed to finally create the Elixir of Life. With the elixir completed, Rien prepares to set off to Japan to begin her plan of creating more of the elixir but is interrupted by the arrival of a second landing party. The elixir was soon stolen by Shija during their encounter with Rien and was later handed to Shugen who secretly drank a small sample of the elixir to check for any hidden poisons before the time came to give it to his superiors.
Departure Arc[]
The Elixir of Life was later returned to Rien after the Tensen defeated Shugen on her ship. Upon his confrontation with Gantetsusai, Shugen felt the effects of the elixir affecting his body, which healed his severed arm. After Rien's defeat, the Elixir of Life was retrieved by the surviving humans who sailed back to Edo to hand it over to the shogun. Before the ruler consumed it, the authorities had the leader of Iwagakure forcibly taste test the elixir for any ill side effects as punishment for an illegal action against the shogun. As a result of the chief being transformed into a tree, Aoki doubted that the elixir would be beneficial to the shogun but was convinced by Jikka that the shogun would viewed as a tree that was divine in exchange for his humanity.

- Since the full effects of the Elixir of Life have not been shown by someone who fully drank it and survived its Arborification process, it is unknown if the elixir lives up to its reputation, as Yamada Asaemon Shugen had only acquired a low healing factor after tasting only a small portion of the Elixir of Life and died upon using up all of his remaining Tao. It is also unclear as to how the Elixir of Life differs from its prototype in terms of ingredients or creation process.
